Bonavera Count 5R

Anemia is a big threat worldwide. According to a report published in a fortnightly magazine from Delhi Down To Earth in December 2020, as many as 68.4 percent children and 66.4 percent women surveyed, suffered from anemia in 2019. These figures were 35.7 percent and 46.1 percent respectively in 2016.

The four most essential steps for anemia diagnosis and classification include:

  • CBC test,
  • Reticulocyte count,
  • Biochemistry, and
  • Bone marrow.

Amongst these tests, reticulocyte counting is one of the most important tests as the number of reticulocytes in circulating blood reflects the erythropoietic (RBC producing) activity of bone marrow. Moreover, they are also useful for cancer patients since anemia is one of the most frequent side effects of anticancer therapy.

The conventional method for reticulocyte is using light microscopy by staining the smear with supravital dyes like NMB. Counting of 2000 cell is recommended by CLSI H44-A2 guidelines. This is a very time-consuming and labor-intensive method. Moreover, error rates range between 20 and 40 percent as reported by many editorials.

In modern hematology, various methods are employed by analyzers to measure reticulocyte count. This method involves NMB or fluorescent dyes that penetrates the blood cells and binds to RNA or DNA in the nucleated cells. Then forward scatter and fluorescence intensity of each cell is measured. The scatter plots are used to determine populations of matured RBC and reticulocytes.

Bonavera Count 5R is the new artifact, which can measure reticulocyte count along with enumerating five parts of WBC populations. It provides results for 28 parameters including Retic_Abs count, Retic percent, and IRF. The throughput of instrument is 60 samples/hour in CBC mode and approximately 45 samples/hour in the retic mode, and the storage capacity is of 200,000 results including data and graphics.

The instrument incorporates unique laser-scatter base MASS technology for enumeration of all five different populations of WBC, in which scattered light signals received by the photodetector at each angle are converted into pulse signals with different amplitudes. By analyzing the pulse signals of different angles, the scatter diagram is obtained, which represents the cell volume and related information. WBCs are classified by the distribution of the pulse signals and the scatter diagram. Moreover for 5-part differentiation, unlike other instruments Bonavera Count 5R uses sheath-flow reagents, which can help keeping WBC population in its approximate original state, that can generate better scatter signals to ensure precise counting of WBC population.

For reticulocyte counting, it uses the NMB dye. The stained reticulocytes along with RBC then pass through the laser beam, and by using polarized light at 90° the different scattering generated by reticulocytes is used to count reticulocytes.

It is one the most convenient and simplest system for reticulocytes as customer can perform even single test of retics and does not require higher work load for the same. Thus, using the technology comparable to other high-end models, it offers convenience in justifying use for low- to medium-sample work load customers.
